IMPDH2 and HPRT expression and a prognostic significance in preoperative and postoperative patients with osteosarcoma
Osteosarcoom is een van de meest agressieve  bottumoren  bij jongeren en adolescenten. Groei van efficiënte therapeutische keuzes blijft ontbreken vanwege de complexiteit van de genomische achtergrond. In eerder werk hebben we een door proteomics geleide drug gebruikt om mogelijke remedies voor osteosarcoom te ontdekken.
Ons vervolgonderzoek onthulde dat een door de FDA goedgekeurd immunosuppressivum, mycofenolaatmofetil (MMF), gericht op inosine-5′-fosfaatdehydrogenase (IMPDH) -enzymen, een antitumoreffect  heeft dat veelbelovend leek voor aanvullend onderzoek en medische proeven.
Profilering van IMPDH2 en hypoxanthine-guaninefosforibosyltransferase (HPRT), de belangrijkste purine-metaboliserende enzymen, kan het begrip van de betekenis van het purinemetabolisme bij osteosarcoom verdiepen en bewijs leveren voor uitgebreid gebruik van MMF in de kliniek.
Binnen het huidige onderzoek, onderzochten we reeksen van IMPDH2 en HPRT in biopsie van 127 exemplaren en na chemotherapie  weefsels  in 20 gevallen van high-grade osteosarcoom patiënten gebruik te maken van immunohistochemische ( IHC ) evaluatie . Er zijn Cox-regressieanalyses uitgevoerd om de prognostische betekenis van alle enzymen te achterhalen.
De resultaten gaven aan dat lage HPRT-bereiken aanzienlijk zijn gerelateerd aan een overmatig Enneking-stadium (P = 0,023) en gemetastaseerd staan ​​(P = 0,024). Univariate en multivariate analyses lieten zien dat patiënten met een lage HPRT-expressie kortere gevallen van totale overleving hebben [HR 1,70 (1,01-2,84), P = 0,044].
Bovendien zijn buitensporige IMPDH2/HPRT-ratio’s in gelijke mate gerelateerd aan kortere gevallen van totale overleving [HR 1,67 (1,02-2,72), P = 0,039]. De reeksen van de enzymen zijn bovendien onderzocht in weefsels na chemotherapie  . De uitkomsten bevestigden dat primaire IMPDH2-expressie gerelateerd was aan kortere metastasevrije overleving [HR 7,42 (1,22-45,06), P = 0,030].
Deze resultaten bevelen een prognostische waarde aan van expressiepatronen van purine-metaboliserende enzymen voor het pre- en post-chemotherapie-interval van osteosarcoomremedie.

Kenmerkend voor het moleculaire en immuunpanorama van de hondenblaas  De meeste kankers

Transitional cell carcinoma (TCC), ook wel urotheelcarcinoom, is de meest voorkomende blaas  meeste vormen van kanker  in mensen en honden Ongeveer een kwart van de menselijke TCC zijn spier-invasieve en in verband met een overmatige risico van verlies van het leven van metastase.
Canine TCC (cTCC)  tumoren  zijn meestal hoogwaardig en spierinvasief . Gedeelde overeenkomsten in gevaar elementen , histopathologie en medische presentatie raden dat CTCC kon functioneren van een mannequin voor de evaluatie van nieuwe therapieën die zal therapieën informeren voor menselijke spierinvasieve TCC.
Het doel van dit onderzoek was om cTCC te karakteriseren op de moleculaire graad om de drijvende krachten achter oncogenese en druggable targets te bepalen. We voerden volledige exome-sequencing (WES) uit van 11 cTCC-  tumoren en drie gematchte reguliere monsters, waarbij we 583 varianten in eiwitcoderende genen uitvonden .
De meest voorkomende variant was een V-naar-E missense-mutatie in BRAF, herkend in vier van de 11 monsters (36%) via WES. Sanger-sequencing herkende BRAF-varianten in acht van de identieke 11 cTCC-monsters, naast in 22 van de 32 in formaline gefixeerde paraffine ingebedde (FFPE) cTCC-monsters, wat wijst op een totale prevalentie van 70%. RNA-Seq werd uitgevoerd bijpassende de genexpressieprofielen van CTCC  tumoren  met regelmatige blaas  weefsel .
cTCC-  tumoren  vertoonden opwaartse regulatie van genen die betrokken zijn in de celcyclus, DNA- herstel en antivirale immuniteit. We bovendien analyseerde het immuunsysteem panorama van CTCC gebruik te maken van het immuunsysteem gen handtekeningen en immunohistochemische ( IHC evaluatie . Een subset van  tumoren  had kenmerken van een verschroeiende tumormicro-  omgeving (TME) en vertoonde overmatige expressie van handtekeningen gerelateerd aan volledige respons op PD-1/PD-L1-blokkade in de meeste kankers van de menselijke blaas  .
Gemultiplexte digitale ruimtelijke profilering van invasieve borsttumoren  van zwarte en blanke meisjes
De NanoString GeoMx Digital Spatial Profiling (DSP) is een gloednieuw gemultiplext platform dat de overvloed aan tumor- en immuungerelateerde eiwitten kwantificeert  in een ruimtelijk opgeloste methode . We uitgevoerd DSP voor het gelijktijdig evalueren van 52 analyten in ruimtelijk gescheiden  weefsel  compartimenten geschetst door pan-cytokeratine expressie.
We hebben daarentegen eiwitdoelen tussen 94 Afro-Amerikaanse / zwarte en 65 Europese Amerikaanse / blanke gevallen ,  tumor-  en stromale  weefselcompartimenten  , oestrogeenreceptor-alfa (ER) -positieve en ER-negatieve instanties en onderzochten mogelijke biomarkers voor overleving. Van 33 analyten met stevige teken voor de evaluatie de resultaten zijn zeer repliceerbaar.
Voor een subset van markers onthulden correlatieve analyses tussen DSP-analyten en conventionele immunohistochemie ( IHC ) scores redelijke tot zeer robuuste associaties tussen de 2 platforms. Eveneens , DSP analyten en scores genexpressie zijn concordante 21 van de 25 merkers met overlap tussen de 2 datasets.
Een aantal analyten die uiteenliepen van ER- stand , en van de 25 onderzochte immuunmarkers, hadden 14 een grote inverse verwantschap met ER-expressie. B7-homoloog 3 (B7-H3; gecodeerd door CD276) was de enige analyt die op een groot onderscheid naar ras wees, namelijk afname in elk van de  tumor-  en stromale compartimenten bij zwarte meisjes . DSP markers die zijn gerelateerd aan de overleving opgenomen CD8, CD25, CD56, CD127, EpCAM, ER, Ki-67, en STING.
We concluderen dat DSP een milieuvriendelijk apparaat is voor het screenen van  tumor- en immuungerelateerde markers in een gelijktijdige mode en resultaten oplevert die in overeenstemming kunnen zijn met gevestigde immuunprofileringstests. DSP immuun te analyseren zijn omgekeerd gerelateerd aan ER expressie, in schikking met een grote lichaamsbouw van het eerdere werk dat papierwerk toegenomen immuun infiltratie in ER-negatieve borstkanker  kanker .
Deze ervaring is gebleken dat de scores van de B7-H3 eiwit zijn geweest aanzienlijk afnemen in de borst  kanker  van Black meisjes in contrast met witte meisjes , een intrigerende ontdekking dat de replicatie in vereist onpartijdige en raciaal talrijke vrouwelijke bevolking.
Terugkerende kinderachtige inflammatoire myofibroblastische  tumor  van mesenterium – casusrapport en overzicht van beeldvormingsbevindingen
Inflammatoire myofibroblastische tumoren  (IMT) zijn ongewoon delicate weefsel tumoren  van intermediaire maligne potentie met een neiging voor inheemse herhaling. Hoewel ze zullen gebeuren in enig opzicht leeftijd teams , prevalentie bij zuigelingen is buitengewoon ongewoon en hun imaging traits mag niet goed beschreven. Een 3-maanden oude vrouwelijke peuter geïntroduceerd met regelmatig progressieve maag uitzetting met niemand   koorts of gewichtsvermindering .

Ze had een grote, slecht gedefinieerde homogene hypodense laesie van 8,4 × 11,4 × 11,3 cm (APxTraxSag) in de maag en vertoonde een lichte verbetering van het onderscheid bij vertraagde verzending. Ze onderging een verkennende laparotomie met totale excisie van de mesenteriale massa, waarvan de histopathologie duidde op IMT.
Ze had een recidief binnen 6 maanden van volledige resectie met een goed gedefinieerde heterogeen versterkende laesie met een afmeting van 1,8 × 1,8 × 2,3 cm (APxTraxSag) in het juiste paravesicale gebied dat tegen de blaas aanligt zonder invasie met een analoge laesie met een afmeting van 4,4 × 2,1 × drie cm (APxTraxSag) in het linker subdiafragmatische gebied dat grenst aan de bovenste bodem van de milt (geen invasie).
Aangezien de chirurgische ingreep bij onze aangedane persoon splenectomie en gedeeltelijke cystectomie met zich mee zou brengen, was voor haar een systemische remedie met ceritinib (anaplastisch lymfoomkinase [ALK]-remmer) bedoeld, waarmee ze na 2 maanden een bijna volledige respons had.
