Immuuncheckpointremmers (ICI) hebben hun werkzaamheid aangetoond bij de behandeling van sterke kankers . Desalniettemin is er geen uniforme voorspellende biomarker voor ICI’s. We wilden het prognostische effect controleren van het gebruik van drie PD-L1-antilichamen (SP142, SP263 en 22C3) voor immunohistochemische ( IHC ) evaluatie.
We hebben retrospectief tumorweefsels onderzocht die zijn afgeleid van 316 omstandigheden van de meeste borstkankers , door weefselmicroarrays vast te stellen en door IHC- kleuring uit te voeren. De immuuncelexpressielading (voor SP142 en SP263) en de gemengde proportionele beoordeling (voor 22C3) waren geëvalueerd en overlevingsresultaten waren geanalyseerd.
Er waren voorspellingsmodes ontwikkeld en waarden van Harrel’s c-index en gebieden onder curven waren berekend om de discriminerende energie te controleren. Ongunstige PD-L1-expressie, voornamelijk gebaseerd op de 22C3- IHC- assay, werd besloten als een onpartijdige prognostische marker voor recidiefvrije overleving (RFS, P = 0,0337) en metastasevrije overleving op afstand (DMFS, P = 0,0131).
Desalniettemin onthulde PD-L1-expressie, voornamelijk gebaseerd op SP142- en SP263- IHC- assays, geen prognostisch effect. Van de vele drie antilichamen, waaronder PD-L1-expressiekennis verkregen via 22C3- IHC- assay tegen de nul-etalagepop, leidde dit tot een grote verbetering binnen de discriminerende energie van RFS en DMFS. We adviseren dat PD-L1-expressie die voornamelijk is gebaseerd op de 22C3- IHC- assay een superieure prognostische marker is dan die welke voornamelijk is gebaseerd op SP142- en SP263- IHC- assays.
Interferon-alfa-induceerbaar eiwit 27 (IFI27) is een prognostische marker voor de meeste pancreaskankers, voornamelijk gebaseerd op volledige bio-informatica-evaluatie
Correct biomarkers te voorspellen het ontstaan en de ontwikkeling van de pancreas adenocarcinoom (paad) worden gezocht in de strijd in de richting van deze dodelijke ziekte . Hier , we gemengde een aantal datasets (GEO, TCGA en GTEX) is het verrichten volledige evaluatie van de alvleesklier meeste vormen van kanker .
Bij wijze van diepgaande evaluatie vonden we dat de expressie van het gen dat codeert voor interferon alfa-induceerbaar eiwit 27 (IFI27) aanzienlijk groter was in de meeste kankerweefsels van de alvleesklier dan in reguliere weefsels , en dat grotere expressie van IFI27 negatief gecorreleerd was met de algemene overleving lading van de alvleesklier meeste vormen van kanker patiënten .
De bruikbare annotatie van IFI27 toonde relaties aan met mobiele immuniteit en metabolisme, met name glycolyse. Evaluatie van infiltrerende immuuncellen weergegeven dat grotere expressie van IFI27 expressie correleert met een verminderde CD8 + T-cellen en verhoogde M2 macrofagen in de tumor immune micro (TIME), dan biochemische analyses van een muis etalagepop en immunohistochemische ( IHC ) kleuring gecontroleerd of glycolytische enzymen en M2-macrofagen aanzienlijk verhoogd in de meeste pancreaskankers weefsels .
We speculeren dat IFI27 kan een effect hebben op de tumor micro-omgeving (TME) van paad door het reguleren van de mobiele immuniteit en de stofwisseling, waardoor de verkoop van de ontwikkeling van de pancreas carcinoom en een verslechtering van de prognose. Deze bevindingen van ons huidige onderzoek zijn een sterk bewijs dat IFI27 een mogelijke prognostische biomarker is van de meeste pancreaskankers en dat het de immuunmicro- omgeving van de tumor beïnvloedt .
Validatie van immunohistochemie voor hondenproteïnen die betrokken zijn bij de opname van jodium in de schildklier en hun expressie in folliculaire celschildkliercarcinomen van honden (FTC’s) en van FTC afgeleide organoïden
Thyrotropinereceptor (TSHR), natriumjodide-symporter (NIS), pendrin en schildklierperoxidase (TPO) zijn belangrijk voor de opname van jodium door folliculaire schildkliercellen. Het doel van het onderzoek was te vergewissen immunohistochemie ( IHC ) protocollen voor TSH receptor, NIS, Pendrin en TPO canine weefsels en karakteriseren van de expressie in organoids afgeleid van honden folliculaire schildkliercarcinoom (FTC) en binnen de bijbehorende grote tumoren .
Dit vormt een elementaire stap om organoïden vast te stellen als een mannequin om de opname van jodium in FTC bij honden te controleren . Commercieel daar antilichamen aan humane eiwitten werden gekozen . De specificiteit van antilichamen werd bevestigd door western blot met gebruikmaking van lysaten van de menselijke schildkliercellijn HTori- Three en een gezonde schildklier van de hond.
IHC werd gevalideerd met behulp van HTori- Drie cellen en een set van honden regelmatig weefsels samen met gezonde schildklier. De expressie van TSHR, NIS, pendrin en TPO werd geëvalueerd in drie organoïde stammen afgeleid van FTC en respectieve grote tumoren .
Alle vier antilichamen produceerden specifieke banden door western blot en cytoplasmatische labeling in folliculaire cellen door IHC in elke menselijke HTori- Three- cellen en de schildklier van de hond. NIS bovendien bevestigd basolaterale membraan immunokleuring in folliculaire cellen. Alle vier de eiwitten waren extreem tot expressie gebracht in organoïden afgeleid van FTC.
De expressie was vergelijkbaar of groter in vergelijking met de eerste tumoren . De resultaten van deze onderzoeken zijn kenmerkend organoids afgeleid van honden FTC als een aanvaardbaar in vitro etalagepop onderzoek jodium opname, het openen van nieuwe analyse perspectieven binnen het onderwerp van canine schildklier kanker geneesmiddel .
Invloed van tumor -infiltrating immuuncellen op inheemse beheer lading , metastasen op afstand, en op overleving bij patiënten met comfortabele tissue sarcoom
Smooth weefsel sarcomen (STS) worden gedacht-over niet-immunogeen, hoewel afzonderlijke entiteiten reageren op anti- tumor brokers richten op de tumor micro-omgeving. Dit onderzoek ‘s goals was geweest om het onderzoek relaties tussen tumor -infiltrating immuuncellen en patiënt / tumor gerelateerde componenten , en hun prognostische beoordelen waard voor inheemse herhaling (LR), metastasen op afstand (DM), en de algemene overleven (OS).
Honderdachtentachtig STS-patiënten (87 vrouwen [46,3%]; mediane leeftijd: 62,5 jaar) waren retrospectief geanalyseerd. Weefselmicroarrays (in totaal 1266 kernen) waren gekleurd met multiplex immunohistochemie en geanalyseerd met multispectrale beeldvorming.
Zeven celvariëteiten waren gedifferentieerd op basis van merkerprofielen (CD3+, CD3+ CD4+ helper, CD3+ CD8+ cytotoxische, CD3+ CD4+ CD45RO+ helper reminiscentie , CD3+ CD8+ CD45RO+ cytotoxische reminiscentie T-cellen; CD20+ B-cellen; CD68+ macrofagen).
Correlaties tussen fenotype-abundantie en variabelen waren geanalyseerd. Uni- en multivariate Mooi & Gray en Cox-regressie mode was gebouwd aan onderzoek prognostische variabelen. Mannequin kalibratie werd beoordeeld met C-index. IHC -findings was gevalideerd TCGA-SARC genexpressie kennis van genen bijzonder voor macrofagen, T- en B-cellen.
Het aandeel B-cellen was lager bij patiënten ouder dan 62,5 jaar ( p = .013), terwijl het aandeel macrofagen groter was ( p = .002). Overmatige B-cel ( p = .035) en macrofaagbereiken ( p = .003) waren gerelateerd aan een verhoogd LR-risico binnen de univariate evaluatie.
Binnen de multivariate setting waren buitensporige macrofaagbereiken ( p = .014) gerelateerd aan een verhoogd LR-risico, ongeacht marges, leeftijd, geslacht of B-cellen. Verschillende immuuncellen waren niet gerelateerd aan het eindresultaat. Buitensporige macrofaagbereiken waren een slechte prognostische kwestie voor LR, ongeacht marges, B-cellen, geslacht en leeftijd. Zo zouden anti- tumor , macrofaag-targeting brokers ook extra regelmatig kunnen worden gebruikt in tumoren met verbeterde macrofaag-infiltratie.
Neoadjuvante pembrolizumab en te hoge dosis interferon-alfa-2b bij reseceerbaar regionaal superieur melanoom
Doelstelling Neoadjuvant immunotherapie kan verbeteren het wetenschappelijk eindresultaat van regionaal superieure bedienbare melanoom en vergunningen voor de snelle wetenschappelijke en pathologische evaluatie van de respons. We onderzochten neoadjuvante pembrolizumab en overmatige dosis interferon-alfa-2b (HDI) oplossing in patiënten met resectabel superieure melanoom.
Lijders en strategieën Patiënten met een reseceerbaar stadium III/IV melanoom waren behandeld met gelijktijdige behandeling met pembrolizumab 200 mg intraveneus (IV) elke drie weken en HDI 20 ME/m2/dag IV, 5 dagen/week gedurende vier weken, daarna 10 ME/m2/ dag subcutaan Drie dagen/week gedurende twee weken. Definitieve chirurgische procedure aangenomen , en adjuvante mengsel immunotherapie die een jaar van remedie afrondt .
Het belangrijkste eindpunt was de veiligheid van de mix . Secundaire eindpunten omvatten algemene respons lading (ORR), pathologische volledige respons (PCR), ziektevrije overleving (RFS), en de algemene overleving (OS). Bloedmonsters voor correlatieve onderzoek waren verzameld allen door . Tumor weefsel werd bepaald door immunohistochemie ( IHC ) en circuleren cytometrie bij aanvang en bij chirurgische procedure .
Resultaten 31 patiënten waren ingeschreven, en 30 waren evalueerbaar. Bij het afsluiten van kennis (2 oktober 2019) was de mediane follow-up voor OS 37,87 maanden ( variatie ; 33,2-43,47). Mediaan OS en RFS zijn niet bereikt. Radiografische ORR was 73,3% (95% CI: 55,5% -85,8%), met een 43% (95% CI: 27,3% -60,1%) pathologische volledige respons (pCR) lading . Geen van de patiënten met een pCR is teruggekomen.

Bij respectievelijk 73% en 43% van de patiënten waren HDI en pembrolizumab stopgezet. Correlatieve analyses stuurden aan dat intratumorale PD-1/PD-L1-interactie en HLA-DR-expressie gerelateerd zijn aan pCR (respectievelijk p=0,002 en p=0,008). Conclusies Neoadjuvante gelijktijdige HDI en pembrolizumab lieten veelbelovende wetenschappelijke oefeningen zien, ongeacht de buitensporige kosten van stopzetting van de behandeling. pCR is een prognostische indicator.
